Discovery and Characterization of an Isopeptidase That Linearizes Lasso Peptides

Lasso peptideThe Link lab has used bioinformatic techniques to find a new class of lasso peptides in water-dwelling alpha-proteobacteria.  A defining characteristic of the gene clusters that encode these lasso peptides is a gene encoding a lasso peptide isopeptidase, a novel enzyme discovered in the Link lab.  Lasso peptide isopeptidase recognizes the specific knotted structure of a lasso peptide and cleaves it into a linear peptide.  The x-ray crystal structure of the enzyme is shown here in a cartoon representation with lasso peptide substrate shown as sticks.
