Sch., Visualizing transcription with single nucleotide resolution using nascent transcript sequencing http://genetics.med.harvard.edu/churchman/ It is now clear that transcription is more complicated than what was thought even a few years ago both in the intricate use of post-initiation control and the mass production of rapidly degraded transcripts. Here we present an approach, native elongating transcript sequencing - NET-seq, that provides a quantitative measure of RNAP density genome-wide with single nucleotide precision. Application of NET-seq in Saccharomyces cerevisiae reveals pervasive polymerase pausing and backtracking and the widespread production of unstable transcripts.
